生物学的媒質を含有するAChEアッセイに対するDIUTHAMEの適用可能性
DIUTHAME (desorption ionization using through hole alumina membrane), a matrix-free laser desorption ionization method, simplifies acetylcholinesterase (AChE) assay measurement and can obtain the relative intensities of the substrate acetylcholine and the product choline with high reproducibility. Previously, DIUTHAME was evaluated for AChE assay using the solutions prepared with Tris-Cl (pH 7.5) 40 mM aqueous buffer. In general, implementation of this assay would involve the use of a relevant biological medium such as blood-derived products. The addition of a biological medium will impact the performance of this assay due to more noise in low mass region as well as ionization suppression. In order to evaluate the feasibility of DIUTHAME for AChE assay involving biological media, this study conducted the comparison of DIUTHAME and MALDI measurements for samples in which sheep serum or raw bovine milk were added.
