酸化ホスファチジルコリンの包括的分析および可視化法
Oxidized phospholipids (oxPLs) play critical roles in numerous pathological events, including cell death and inflammation. However, due to the lack of its structural information and analytical methods, the quantity and pathological behavior of endogenous oxPL species remains unknown. Here, we constructed a library of 465 oxidized phosphatidylcholines (oxPCs) by mass spectrometry (MS)-based nontargeted analysis. Using the developed library, a total of 70 oxPCs were detected in acute liver failure (ALF) induced by acetaminophen (APAP). In particular, doubly-oxygenated PUFA-PCs (PUFA+O2-PCs) containing epoxy-hydroxide group remarkably generated in early phase of liver injury. Furthermore, by coupling in vivo 18O2 labeling and matrix-assisted laser desorption/ionization-tandem MS imaging (MALDI/MS2/MSI), we found the specific accumulation of PUFA+O2-PCs in CYP2E1 expressing hepatocytes, major injured areas of APAP induced ALF. Our developed library and visualization methodology will accelerate research and development in the field of LPO related diseases.
