膜内切断プロテアーゼのリガンド-膜タンパク質複合体のネイティブ質量分析
Native mass spectrometry is attracting interest as a useful analytical tool for membrane proteins. However, native mass spectrometry of membrane proteins is extremely difficult. In this study, we investigated native mass spectrometry for observing membrane protein-ligand complexes using the intramembrane-cleaving protease from Escherichia (E.) coli, for which Compound A is known as a specific inhibitor. Furthermore, using E. coli intramembrane-cleaving protease and two species of orthologs for which specific inhibitors have not been identified, we observed the binding state of three different species of intramembrane-cleaving proteases and the Compound A, and attempted to make a quantitative comparison.
