臨床診断を目的としたキラルアミノ酸の多次元HPLC法開発とヒト血液・尿中における含量解析
The relationship between the renal disorder and D-amino acids has been reported, and D-amino acids are gathering attention as novel biomarkers for the kidney function. Since intrinsic D-amino acids are present in trace level with huge amounts of interfering compounds, a highly-sensitive and selective analytical method is required. In the present study, a three-dimensional HPLC system and a two-dimensional LC-MS/MS system have been developed. Amino acids in the human plasma and urine were pre-column derivatized with 4-fluoro-7-nitro-2,1,3-benzoxadiazole. By using the methods, chiral amino acids in the clinical samples were clearly observed without interferences, and the %D values of several amino acids in the plasma of patients with chronic kidney disease altered along with the progress of kidney dysfunction. The developed methods have high selectivity, and further applications to various clinical samples are expected.
