走査型プローブエレクトロスプレーイオン化法による生体多次元化学分布情報計測
The technology to measure the molecular information in cells and to extract features that express the state of cells is extremely important for the basic understanding of the field of life chemistry and for the advancement of the investigation of diseases. Mass spectrometry imaging (MSI) can visualize the distribution of various biological components by ionizing a group of molecules contained in a biological sample. We have been developing the analytical technique called tapping mode scanning probe electrospray ionization (t-SPESI) for rapid local extraction and ionization under atmospheric condition. In order to perform MSI with high spatial resolution, we developed a measurement method of probe vibration and a feedback control system for SPESI. By irradiating a laser beam from the side of the probe, the vibration amplitude is measured, and the vibration amplitude is kept constant by rapidly adjusting the height of the sample stage. We realized the stabilization of the probe scanning for an uneven sample and achieved high spatial resolution MSI. We will report the development of the t-SPESI system and the results of the measurement of multidimensional chemical distribution information of human heart disease tissue.
