LC/MSを基盤とした血漿メタボローム分析法の性能評価

Metabolomics involves the unbiased quantitative and qualitative analysis of the complete set of metabolites present in cells, body fluids, and tissues. Chromatography coupled with mass spectrometry (MS) techniques are the most commonly used in metabolomics. One of the current problems in the field of metabolomics is the difficulty in integrating data collected using different equipment at different facilities, because many metabolomic methods have been developed independently and are unique to each laboratory. It is suggested that each chromatographic mass spectrometry method has different performances such as the number of detectable compounds, quantitative performance, and matrix effect. Herein, we evaluated the performance of four types of liquid chromatography mass spectrometry (LC/MS), including hydrophilic-interaction LC/MS (HILIC/MS), ion chromatography with an anion exchange column MS (AEX/MS), LC with a pentafluorophenylpropyl column MS (PFPP-LC/MS), and hydrophilic-interaction/anion-exchange LC/MS (HILIC/AEX/MS), for hydrophilic metabolome analysis of human plasma. Our study revealed that PFPP-LC/MS and HILIC/AEX/MS have excellent performance in terms of coverage, repeatability, and quantification of the detected hydrophilic metabolites.
