MALDI-MSを用いたDNA-ペプチド間で生じる相互作用の評価
Recently, oligo nucleotide therapeutics, a type of medicine based on DNA structure, have been developed. X-ray diffraction and NMR were mostly used to evaluate the DNA-peptide interaction, which requires few nanomoles of sample. When mass spectrometry is used only picomole order of sample is required, making it more efficient method to evaluate the interaction. MALDI mass spectra of mixed solution of DNA and peptide, both with equal mole, gave peaks of DNA dimer, DNA-peptide complex and peptide dimer. Dominant peaks differed by peptide sample used. RRL13, peptide containing three arginine residues, gave DNA-peptide complex as a dominant peak, while Chromogranin A, peptide containing one arginine residue, gave DNA dimer as a dominant peak. These results suggested that the salt bridge interaction between phosphate group of DNA and arginine amino acid can be formed.
