Characterization of Antibody-Drug Conjugate Synthesized by CCAP Method

To demonstrate liquid chromatography mass spectrometry (LC/MS) characterization of site-specific Antibody-drug conjugate (ADC) synthesized using chemical conjugation by affinity peptide (CCAP). Conjugation was confirmed by UltraPerformance LC (UPLC) equipped with quadrupole time-of-flight (Q-Tof) MS and drug-to-antibody ratio (DAR) of 2 was calculated by mass shift. Conjugate site was estimated by LC/MS peptide mapping using various enzyme digestion.
