Unraveling Pathogenesis of Renal Amyloidosis with an Integrated Workflow of MALDI Imaging Mass Spectrometry and LC-TIMS-MS/MS on Formalin Fixed Renal Biopsy Tissue Section

Here we have established an integrated workflow of matrix-assisted laser desorption/ionization imaging mass spectrometry (MALDI-IMS) in combination with shotgun proteomics using liquid chromatography, trapped ion mobility and mass spectrometry (LC-TIMS-MS/MS). An analytical technique was developed and evaluated through a combination of specific sample preparation/extraction protocols based on the same formalin-fixed tissue section of renal biopsy tissue samples. Our strategy succeeded in the discrimination of subgroups of renal amyloidosis such as immunoglobulin light chain (AL) and serum amyloid A (SAA) type. With the current strategy, we have detected some of the proteins that may provide insights into the pathogenic process of amyloid formation in human kidneys.
