Rapid and Secure Analysis of in-vitro DNA Methylation by ESI-TOF-MS
We have developed a rapid and secure method using nanoESI-TOF-MS for analysis of in vitro CpG methylation of DNA with <500 bp length. The most commonly used method for confirmation of DNA methylation is gel electrophoresis of the digest of methylation-sensitive restriction enzyme, but this cannot straightforwardly be used for any DNA fragments; DNA sequences that contain CpG sites should be designed to match the specificity of restriction enzyme. ESI-MS can be alternative but thorough desalt is required especially for DNA samples. By optimizing the sample preparation methods and measurement conditions, we have succeeded in analysis of methylation levels of DNA with any sequence using nanoESI-TOF-MS.
