MALDI-MS Proteotyping Based on the Size Select Proteomics

MALDI-MS have been used as a tool for bacteria identification at the genus and species levels especially in the field of clinical microbiology. But, discrimination of bacterial strains intra species is crucially important, because diversity of characteristics such as pathogenicity and immune response. We have proposed bacterial typing method based on MALDI-MS proteotyping in which use ribosomal proteins as biomarkers. However, detection of ribosomal proteins is not always easy due to several bacterial constitutions. Although purification of ribosomal proteins by ultra-centrifugation for at least one hour is effective to detect ribosomal proteins, it is not suitable for practical use. In this study, we developed rapid and simple purification technique of ribosomal proteins for MALDI-MS proteotyping.
