Analysis of Gram-Negative Bacteria Component Lipid A Using LC-QTOF-MS
A method for the analysis of lipid A, an important immune activator of gram-negative bacteria, was developed by using LC-QTOF-MS. The sharp peak of lipid A was observed by employing a metal-free column and EDTA-containing solvent. MS/MS spectra assignment enabled the structure estimation of lipid A. This method was successfully applied to the analysis of cultured Escherichia coli in vitro and fecal sample of wild type mice. The results suggest the structural diversity of lipid A in fecal microbiome.
