Localization Analysis of Catecholamine via Mass Spectrometric Imaging

We investigated the localization and metabolomics of catechol amines such as L-DOPA, dopamine (DA) and norepinephirine (NE) in which engage physiological change of mice brain. 2,4,6-trimethylpyrylium tetrafluoroborate (Py-I) can selectively and efficiently react with the amino group by the reaction of pyrylium salts with nucleophiles under alkaline conditions, thereby increasing the mass by 105.1. Py-I-labeled L-DOPA, DA and NE showed m/z 302.2, 258.1 and 274.1 increasing the mass by 105.1 from original mass, respectively. MSI showed the change of localization of target molecules. L-DOPA and DA localized at brain stem and SN, and ventral striatum.
