Influence of Solvent Composition on the Threshold of Laser Fluence in MALDI
In Matrix-assisted laser desorption/ionization (MALDI), nitrogen pulse laser photons are irradiated to the crystal surface of matrix and analyte, and then the surface is ablated while matrix and analyte molecules are ionized in the plume. The matrix crystal shape and threshold laser fluence depend on various factors such as matrix, analyte and solvent composition. The size of crystal of α-cyano-4-hydroxycinnamic acid(CHCA), one of typical matrices of MALDI, decreased with increasing the ratio of acetonitrile in mixed solvent of water and acetonitrile. Threshold laser fluence of ion generation dropped with decreasing in size of crystal. Other matrixes (sinapinic acid and 2,5-dihydroxybenzoic acid) were used in the same way as CHCA.
