Development of Chiral-Separation Analytical Method of α-Hydroxycarboxylic Acid Metabolites for Use in Disease Marker Analysis
We developed a chiral-separation analytical method of α-hydroxycarbolic acid metabolites. 2-Hydroxyglutaric acid (2-HGA) is a chiral α-hydroxycarbolic acid metabolite, and it is reported as a cancer-specific biomarker. After acetylation of the α-hydroxy group in the structure of 2-HGA, good enantioseparation was achieved by performing diastereomeric derivatization with DMT-3(S)-Apy. From quantification results of D- and L-2-HGA in saliva samples obtained from healthy persons and cancer patients, an increase of in D-2-HGA was confirmed in the patient groups, suggesting that the developed method would be usable in clinical diagnosis.
