Improvement of Sampling Ionization Efficiency in Tapping-Mode Scanning Probe Electrospray Ionization (t-SPESI)

Ambient sampling and ionization (ASI) technique have been attracted attention not only for the rapid and direct analysis but also for the imaging mass spectrometry. Tapping-mode scanning probe electrospray ionization (t-SPESI) realizes ASI with a single capillary probe. The formation of a liquid bridge and an electrospray at a vibrating capillary probe apex are utilized for the sampling and ionization, respectively.
In order to realize the reproducible high spatial resolution and ionization efficiency by t-SPESI, we studied the effect of experimental parameters onto the formation of liquid bridge and the ionization results. The parameters such as vibration frequency of capillary probe, solvent voltage and flow rate are considered in the present study. This is because the vibration frequency is considered to modulate the time interval between the liquid bridge formation and the electrospray generation. The solvent voltage is also considered to change the sampling spot size due to the accumulated charge in a liquid bridge. The solvent flow rate determines the total volume of solvent supplied from the probe apex. This study is essentially important not only to understanding the dynamic change of liquid bridge but also to control the sampling spot for the improved t-SPESI.
