Development of Highly Sensitivity Method in LC/MS/MS Analysis of Peptides Using Ammonia Eluent
We compared the effects of formic acid and ammonia on the elution of four modified peptides. The peak shape and signal to noise of the phosphorylated peptide were improved by using a metal free column. This improvements are due to the reduced adsorption of metals in the column. The sensitivity of the three peptides was improved 2 to 3 times by using ammonia eluent. In addition, the peak sharp of the phosphorylated peptide was improved. The ammonia eluent was reduced the interaction between the phosphate group and the metals.
