Imaging Mass Spectrometry of Lipids

We have demostrated the significance of lipids concerning development of disease and/or living body using imaging mass spectrometry (IMS). A number of ionization methods have developed in IMS, the most commonly employed to date being matrix-assisted laser/desorption ionization (MALDI). Additionally, desorption electrospray ionization (DESI) and secondary ion mass spectrometry (SIMS) were performed in accordance with the purpose.
MALDI-IMS has been applied to the analysis of lipids and other molecules. We showed that some phospholipids were increased in cancer samples and fourier transform ion cyclotron resonance MALDI-IMS which provide the high mass resolution compared to time-of-flight MALDI-IMS revealed that there were 194 peaks with m/z difference of less than 0.2 in rat brain.
DESI-IMS has been applied to the detection of free fatty acid and other lipid species. We performed DESI-IMS study of aorta in ApoE-deficient mice were fed containing EPA or DHA. DESI-IMS showed that not only EPA and DHA but also each metabolites were accumulated in plaques.
SIMS-IMS is a technique used to analyze the composition of solid surfaces. We tried to intercellular lipid imaging by pre-treatment of biological samples.
This tutorial presentation mainly focused reviews the different techniques and methodologies employed for 3 types of IMS.
