S-Trap法によるHeLa細胞の膜タンパク質LC-MSプロテオーム解析

In this study, the applicability of S-trap method is proved. In the S-trap method, all the proteins in the cells are solubilized by high concentration of SDS (5 %) containing Lysate buffer treatment and the degraded proteins are trapped in the S-trap spin column. Sequentially, the trapped protein is digested to peptide size by enzymes on the spin column. This method showed high digestion efficiency, especially, trypsin digestion was completed in 1 hour at 47 C. Whole enzymatic digestion procedure of protein which normally takes two days is completed in a half of a day and can be subjected to LC-MS assay. The effectiveness of this S-trap method was verified by comparing to existing In-gel digestion method and PTS method.
