血清タンパク質複合体解析によるブタ非アルコール性脂肪肝疾患から肝細胞癌発癌に至るバイオマーカーの同定
Noninvasive biomarkers are urgently needed for optimal management of nonalcoholic fatty liver disease (NAFLD). Here, we established a pig model replicating hepatocellular carcinoma (HCC) associated with NAFLD. Using this model, we performed serum proteomics in order to identify novel biomarkers. Serum proteins were separated by blue native two-dimensional gel electrophoresis in terms of multiple protein complex analysis. By MALDI-TOF MS/MS following separation, serum inter-alpha-trypsin inhibitor heavy chain 4 (ITIH4) was identified as the one of most characteristic protein corresponding with NAFLD progression and HCC development. In immunoassay, serum ITIH4 levels in NAFLD pigs were chronologically increased in comparison with those in control animal. In the validation for human samples, serum ITIH4 levels were significantly higher in NAFLD-HCC patients. Serum ITIH4 promises to have clinical utility in monitoring NAFLD progression and subsequent HCC development.
