Systematic analysis on skin aging caused by intrinsic or extrinsic factors

Intrinsic and extrinsic aging would result in significant alteration to skin structure and components, leading to subsequent impact on molecular cascades and biochemical processes. In this regard, we have performed the immunohistological evaluation and revealed the proteome profiles of nude mice skin to elucidate the specific responses during aging caused by various factors. Our findings indicated that UVB-elicited (extrinsic) aging mainly caused upregulation of PCNA and strong oxidative damage in DNA, whereas chronological (intrinsic) aging diminished epidermal cell growth and increased the expression of caspase-14, as well as protein carbonylation. Moreover,network analysis also showed that the programmed skin aging could trigger the ubiquitin system
and remarkably suppress the level of 14-3-3 sigma, which might imply the loss of cell growth capacity. On the contrary, UVB exposure induced inflammation and the risk of skin carcinogenesis.
Taken together, functional proteomics provides a high-throughput approach to investigate the potent proteins and molecular events which play critical roles in skin subjected to both intrinsic and
extrinsic aging.
