Label-Free Quantitative Mass Spectrometry of Urine to Identify Cervical Cancer Biomarkers

Cervical cancer is one of the most common cancers. Pap smears, the invasive method, is used for diagnosis cervical cancer. There is still a need for searching biomarkers from biological fluid. The comparative proteomics was used to identify potential protein biomarkers in urine samples from normal and cervical cancer patients. Quantitative label-free mass spectrometry analysis was used to perform the urinary proteome differences between normal and cancer patients and the immunodetection was used to confirm the interesting proteins for the potential biomarkers. The urinary proteomes alteration from normal and patients showed 59 and 73 for up-and down-regulated proteins, respectively. The method could detect high molecular weight proteins from 100 kDa when compared to two-dimensional gel electrophoresis technique. The biological pathway involved various functions including the blood coagulation, cell-cell adhesion, metabolic process, regulation of morphogenesis and stress response. The interesting upregulated-urinary proteins was leucine-rich alpha-2-glycoprotein (LRG1) while the down-regulated proteins were S100-A8 (S100A8). The validation of these 2 proteins were confirmed using immunodetection. In conclusion, the quantitative mass-spectrometry-based analysis was used. We found 2 interesting proteins which could be useful to be the potential biomarkers for cervical cancer.
