We would like to introduce the study of enzymes using the small molecular fluorescent substrate probes that can report enzymes' live functions in bio-samples. We have developed the methodology of characterizing disease-related enzymes by globally monitoring their activities using the library of substrates, and discovering and characterizing ones that showed alternation in disease-derived samples. In one example, a global monitoring of the peptide-metabolizing enzymatic activities led to the discovery of novel endopeptidase activity that was elevated in colorectal tumor cells. We expect that the omics of enzyme, enzymomics, will proceed for further characterization of disease-related proteins as potential drug targets or biomarkers.
