Systematic Proteomic Quantification on Atherosclerosis Progression using Knockout APOE Mice Model

Atherosclerosis (AS) is the main cause of coronary heart disease and other cardiovascular diseases that severely threatening human health worldwide. As current treatment strategies of atherogenic hyperlipidemia hyperlipidaemia prevent only 30% of all cardiovascular events, novel treatment strategies are highly warranted. Atherosclerosis is a chronic and progressive multifocal arterial disease associated with lipid deposition, inflammatory response and abnormal remodeling of the arterial wall. The complicated disease process of AS hampered research works from fully uncover the pathological mechanisms.
In this study, we adopted quantitative proteomic analyses on the blood vessels of the APOE-/- and WT mice with normal diet (ND) or high fat diet (HFD). More than 6, 200 proteins have been quantified in total. Through comparing the differential proteins of APOE-/- HFD vs ND and WT HFD vs ND, proteins related to MYOCD inhibition and necrosis activation were highlighted. The variations of protein expression represented the remodeling of the vascular smooth muscle cells (VSMC) during AS progression. Further functional study is ongoing, and this research would pave the way for discovering novel risk factors and treatment strategies.
