Serotonin is produced in both the brain and gut and plays important roles in both organs. This suggests serotonin functions in brain-gut interactions. The relationship between serotonin and dysphoria has been reported. The serotonin content in feces can indicate the serotonin production status in the gut and can be an indicator of dysphoria. The analysis of trace substances in feces is very difficult due to the large amount of contaminants, so pretreatments such as solid phase extraction are needed. Therefore, in developing an analytical method of serotonin in feces, we utilized solid phase extraction and column-switching LC/MS using human feces containing a stable isotope-labeled serotonin. A preliminary result we obtained was a 29.9±3.8 % of recovery rate using three subjects’ feces. The content of endogenous serotonin in their dry feces was 1–2 ng/mg.
