アミド化によるエイコサノイド類の高感度LC-MS/MS分析手法の開発

Eicosanoids play a number of important roles in inflammation/anti-inflammation processes. Detection of these molecules is important for understanding not only disease mechanism but pharmacological and toxicorogical effects of drugs. However, contents of eicosanoids in biological samples are extremely low and it is difficult to detect them from a limited sample with sufficient measurement sensitivity. We have developed a sensitive simultaneous analytical method for eicosanoids by derivatization of carboxylic group in eicosanoids into amide under mild condition by using 4-(4,6-dimethoxy-1,3,5-triazin-2-yl)-4-methylmorpholinium chloride (DMT-MM). This method can detect various eicosanoids from mice bronchoalveolar lavage fluid with high sensitivity and reproducibility.
