がん細胞株のセクレトーム解析を用いた脳腫瘍細胞分泌タンパク質の同定

Biomarker for glioblastoma (GBM) is identified from serum, plasma or cerebrospinal fluid. However, abundant proteins in blood or cerebrospinal fluid interfere identification and quantification of small amount of protein derived from tumor tissues. Therefore, the purpose of the present study was to identify GBM cell specific secreted proteins by analyzing conditioned medium (CM) of GBM cell lines (U87, U251, T98G) and various cancer cell lines (MDA-MB-231, MCF-7, Caco-2) using quantitative proteomics. The CMs were collected and digested by trypsin. Digested peptides were identified by data dependent acquisition (DDA) and quantitated by data independent acquisition (SWATH-MS). As a result, we identified 26,041 peptides derived from 2,371 proteins. Among identified proteins, 21 proteins were selected as candidate proteins, which were specifically secreted from GBM cell line. Then, 25 peptides, of which expression levels were highly correlated with those of corresponding proteins, were selected for MRM analysis. As a result, 19 peptides significantly increased in CM of GBM cell lines compared to the others cell line. In conclusion, we identified proteins that were specifically secreted from GBM cells. These proteins are expected to be candidates of markers or therapeutic targets for GBM.
