精密医療のための定量的プロテオームプロファイリング:基礎研究から大規模解析に向けて

The transition from discovery proteomics to translational research in the pipeline of precision medicine means that we are no longer measured by the number of protein identifications, highest sensitivity or best coverage, but success criteria focused on quantitation, reproducibility, standardization, scalability, access to curated references, and rigorous statistics. These studies require implementation of highly robust analytical platforms to deliver accurate and reproducible peptide/protein quantitation along with high throughput. Ultimately, verification of protein biomarkers requires confident quantitation of low-level proteins in large-cohort studies in a standardized and precise manner.
In this presentation, we will discuss several new technology developments and optimization of each component in the translational proteomics workflow: sample preparation, separations, mass spectrometry, and data analysis. A particular emphasis will be placed on integrating these to enable end-to-end, complete, standardized solutions for the rigors of quantitative proteomics focusing on label-free precursor-based quantitation.
To demonstrate a highly reproducible quantitative analysis of complex proteomes in cancer research, proteomics profiling of paired cancer and control tissue were carried out based the new advances in Orbitrap technology and workflows.
