The need for accurate description of protein behavior and function in solution has gained importance in various fields. To achieve the desired accuracy, multiple precise analyses should be performed on the target molecule with the cutting-edge proteomic approaches. We will present the effective approaches to determine the accurate molar mass value of the target proteins and the mechanism of microtubule (MT) cytoskeleton dynamics, as an example. MTs are dynamic structures that are fundamental for cell morphogenesis and motility. KIF2 depolymerizes MTs during ATP hydrolysis. However, the detailed mechanism by which KIF2 drives MT depolymerization remains unknown. To elucidate the mechanism, the transitional KIF2-tubulin complex was analyzed by multiple approaches: crosslink mass spectrometry, size-exclusion chromatography, light scattering, small-angle X-ray scattering, analytical ultracentrifugation, and atomic force microscopy. The analyses outlined the transitional conformation of KIF2-tubulin complex, and X-link MS determined the absolute molecular value and the critical interaction of the complex.
