定量プロテオミクスによるヒト血清と血漿中タンパク質の比較と安定性の解析

Proteome analysis of human serum and plasma is standard strategy to identify new biomarkers of various diseases. However, the proteins, which expression were changed depending on the store condition, were not suitable for the biomarkers. Therefore, the purpose of this study was to assess the differences in proteome between serum and plasmas, and stability of human plasma proteins under the different store conditions after blood collection by quantitative proteomics. Human serum and plasma which prepared under various conditions was digested with trypsin and analyzed by nanoLC-SWATH-MS. Plasma-rich proteins were fibrinogen and coagulation factor, and serum-rich proteins were platelet factor 4 and platelet basic protein. No protein leaving at room temperature for 15 or 30 minutes changed more than 1.5-fold compared with those for 0 minutes, and 6 proteins leaving on ice for 4 or 8 hours changed more than 1.5-fold compared with those for 1 hour. Among them, Hemoglobin and Carbonic anhydrase 1 gave high peak area in the hemolysis samples. The present results suggest that major part or proteome was not different between plasma and serum, and also stable under the store conditions in the present study.
