Lectin microarray (LMA) is a lectin-assisted glycomic technology useful to assess characteristics of glycan structures with a simple and rapid procedure, facilitating glycoproteomics by applying prior to detailed glycan structural analysis by mass spectrometry-based approaches. In our previous work, by the combined use of laser microdissection (LMD) and LMA, we have established a standardized method of differential glycomic profiling for formalin-fixed paraffin-embedded (FFPE) tissue sections. This LMD-LMA method has been validated to apply to FFPE mouse tissue sections of the whole body, providing comprehensive tissue glycome mapping data to overlook site- and tissue-specific protein glycosylations. For cell type-specific analysis, a possible strategy for more accurately targeted dissection is a direct dissection from tissue sections stained with a specific probe (i.e. antibody and lectin) against a cell type-specific molecular marker. Here, we established the improved LMD-LMA method for a direct analysis of FFPE tissue sections fluorescently stained with a lectin and antibody against an endogenous marker, enabling cell type-specific glycomic analysis of tissue specimens with more accuracy and precision. The present method is expected to apply to differential glycomic analysis of pathological specimens in combination with immunohistochemistry for disease-relevant markers and thus contribute to discovery of disease-related glyco-biomarkers and therapeutic targets.
