イメージング質量分析法を用いたショウジョウバエ体内におけるイミダクロプリド可視化手法の開発

Imidacloprid, which is one of the neonicotinoid insecticides, is widely used all over the world for exterminating harmful insects. While it is known to have agonistic action on nicotinic acetylcholine receptor in insects, there is little information about insecticide distribution in insects. Hence, there is no information on how imidacloprid is distributed after being incorporated. In this research, we developed the visualization method for imidacloprid in Drosophila melanogaster, as a model insect, using matrix-assisted laser desorption/ionization imaging mass spectrometry (MALDI-IMS).
IMS requires sample cryosections, however there are some challenges for obtaining good morphology sections of Drosophila melanogaster because of its small size and heterogeneous components. Therefore, the section preparation method was optimized first, and then imidacloprid distribution in Drosophila melanogaster was visualized using MALDI-IMS.
By using 10% gelatin as an embedding material and 70% ethanol as a pretreatment, the gaps between embedding material and Drosophila melanogaster were reduced, and hence good morphology section could be obtained. Next, imidacloprid standard was analyzed by MALDI and ESI separately, and imidacloprid was found to be changed into guanidine-imidacloprid by MALDI laser irradiation. As a result, imidacloprid distribution in Drosophila melanogaster could be successfully visualized using guanidine-imidacloprid as the target.
