短鎖ペプチド探索の分析フレームワーク: ペプチド予測とタンデム質量分析の統合

Naturally occurring short peptides contribute to a great variety of biological functions, such as neuro transmission, hormone signaling, host defense and many others. Analysis of short peptides, especially exploration of novel functional peptides is of particular importance since it potentially leads to the finding of important biological processes or drug development. However, sensitive and reliable identifications for short peptides are still difficult even with the modern mass spectrometry instruments.
we have developed an analytical framework dedicated to the exploration of short peptides. We created a tool that enables to predict naturally occurring peptides based on a given proteome or genome sequence dataset. This tool crates potential forms of short peptides based on user-defined structural properties such as substructures, motifs, potential cleavage sites, post-translational modification and other molecular properties. Together with top-down MS/MS spectrum matching and similarity matching for in-silico fragment spectra, our method enables highly sensitive short peptide identification, also taking into account unexpected modifications. Our analytical framework is built in a modular together with other informatics, data analysis tools, and therefore is flexible and applicable to any kind of peptides in a wide variety of matrices.
