SWATH-MSとLasso回帰分析を用いた黒毛和種牛血清の経時的プロテオーム解析による枝肉形質バイオマーカー候補タンパク質の探索
The technology for early assessment of carcass traits and meat quality characteristics during fattening is expected to improve the efficiency of beef cattle production. However, there have been few reports on developing an index for evaluating the degree of fattening of Japanese Black cattle. To discover protein biomarkers which can be used to chronologically evaluate individual variations of carcass traits and meat quality characteristics in fattened cattle, we carried out proteomic analysis of serum using SWATH-MS and Lasso regression analysis. Serum samples were collected from Japanese Black steers (n=44) at 13, 15, 17, 19, 21, and 23 months of age. Solubilized serum proteins were digested with trypsin at 37 degrees C. The tryptic digests were desalted with ZipTip uC18 and measured by LC-MS/MS with SWATH acquisition. In SWATH-MS analysis, total of 174 proteins were detected and 135 proteins were selected for quantitative analysis. Several proteins were selected by Lasso regression analysis as explanatory variables against carcass traits and meat quality characteristics. Those results suggest possibilities of selected proteins as protein biomarker candidates for predicting carcass traits and meat quality characteristics during fattening of Japanese Black cattle.
