The cell, the basic biological unit of all organisms, is surrounded by the plasma membrane. the membranes not only serve as barrier to separate the interior of cells from outer environment, it also serves as the field for biological reactions. The membrane contains a variety of molecules, especially proteins and lipids. According to the fluid mosaic model, there would be various structures consist of protein-protein complex and clustered lipids on the membrane, yet actual membrane structures are not fully understood. To reveal the membrane structure, we are developing a compound microscope to obtain both super resolution optical image and mass spectrometry image of the membrane from a same specimen. The imaging mass spectrometry was achieved using stigmatic laser desorption/ionization mass spectrometry to reveal the subcellular distribution of lipid components. The super resolution image is obtained using photoactivated localization microscopy (PALM). PALM imaging revealed the precise distribution of whole copies of receptor protein, which distribute nonrandomly on the membrane.
