MALDI-TOF/TOFを用いる20keV高エネルギーCIDによるペプチド分解の新局面

Positive-ion CID spectra of peptides obtained with 20keV high-energy TOF/TOF MS have been analyzed from the standpoints of selective/enhanced cleavage of Asp/Glu/Cys-Xxx, Xxx-Pro and Gly-Xxx residues by comparing with low-energy CID. The high-energy CID spectrum of a model peptide BSA1-48 containing Asp, Glu, Cys, Pro and Gly residues showed preferential b- and y-ions originated from the cleavage at C-N bond of Asp-Xxx residues, which is a characteristic of low-energy CID. The selective/enhanced cleavage of Asp-Xxx residues in high- and low-energy CID could be explained by assuming the pre-formation of an intramolecular hydrogen-bonding state (Fig. 1left).
