SWATH法による高脂肪食誘導肥満マウスの肝臓の定量プロテオミクス―食品成分の機能性の分子機構解明に向けて―
In this study, we performed SWATH measurements of livers, which is the center of metabolism and deeply involved with the development of lifestyle-related diseases, obtained from high-fat diet (HFD)-induced obese mice, in order to develop the method to elucidate molecular mechanisms of functional food substances. Seven weeks old C57/BL/6J male mice were fed with HFD containing 0.1% cellulose or leave extract of plant A (LE), or normal diet (ND) for 5 weeks. On the day 35, livers were collected after 16 h of fasting. Proteins extracted from the livers were trypsinized, and the resulting peptides were analyzed by SWATH. By the IDA analyses and database-search, 1249 proteins (10880 peptides) were identified. Among them, 760 proteins were successfully quantified. PC-DA of the proteome profiles clearly separated the three groups, indicating that they had the distinctive proteome profiles. Compared to the ND, the amount of many proteins involved in detoxification decreased in the HFD group, which was consistent with previous reports, while the amount of proteins involved in cholesterol metabolism or inflammation increased in the HFD group. The intake of LE affects the abundance of Cytochrome P450 proteins, which may be involved in cholesterol lowering effect of LE.
