全自動プレカラム誘導体化法によるアミン・アミノ酸類の一斉分析と真菌メタボローム解析への応用

Amino Acids (AAs) quantitation is an essential analytical platforms commonly applied in the blood/urine metabolomics studies and early disease diagnosis. However, concurrent analysis and quantitation of AAs need technological efforts in some biological matrix due to its highly hydrophilic and bipolar physicochemical properties, and the lack of specific UV absorption. Pre-column derivatization was contrived as a means of potential and practical solutions by extension of reverse phase LC-MS method. In our study, AAs and amine neurotransmitters were derivatized by 9-fluorenylmethyl chloroformate (FMOC) using the automatic pretreatment function on autosampler in generic HPLC system. Inter-batch analytical reproducibility was improved using isotope-coded internal standards for AAs and several monoamines added before derivatization. The present method covered all kinds of essential and non-essential AAs; and several amine neurotransmitters like histamine (downstream metabolite of histidine), catecholamines (phenylalanine/tyrosine), and serotonin (tryptophan). From the comparative analysis of the plasma taken from ICR mice infected by Candida albicans, the plasma concentrations of methylated amino acids were significantly higher by the infection. Our results suggest that the FMOC auto-precolumn derivatization would be a useful and labor-saving tool for metabolomics/proteomics.
