Concurrent mass spectrometric analysis of multiple samples using Py-Tag reagents

Py-Tag reagents are provided as a set of three stable isotopic reagents. The use of three reagents allows multiplexing of three different samples in a single LC/MS/MS experiment. As model protein, bovine serum albumin (BSA) was digested by trypsin. After digestion, Py-tag-0 (Mw: 175) was added to the solution to modify with lysine residue of fragment peptide. The fragment peptide (m/z 1163) indicated m/z 1338, 1344, 1350 after modification of Py-tag-0. Finally, we compared between normal cell and drug-tolerant cell. After modification of Py-tag of drug-tolerant cell lysate, we investigated the difference of mass between normal cell and drug-tolerance one. Thus, we could easily analyze the difference between control and drug-resistant cells in protein identification and quantitation.
