Simultaneous LC/MS analysis of saccharides, organic acids, and amino acids using polymer-based HILIC column under alkaline condition

In this study, a polymer-based HILIC column Shodex® HILICpak® VG-50, packed with polyvinyl alcohol base material modified with tertiary amino groups, was used to develop high-sensitive LC/MS method for the quantification of saccharides, organic acids, and amino acids. In order to simplify conventional analytical method required derivatization or ion pair reagent, a new LC/MS method was studied. We found that VG-50, with aqueous solution including ammonia / acetonitrile as the eluent, was suitable for this analysis. An alkaline condition (max. pH13) is available for VG-50. It was demonstrated that this method was applicable for the simultaneous analysis of these compounds in a commercial energy drink. This HILIC column was useful for the LC/MS analysis of various hydrophilic substances using more simple conditions than previous methods.
