Investigation of surface-assisted laser desorption/ionization (SALDI) using the silver-coated glass plate for imaging mass spectrometry (IMS)

We investigated the surface-assisted laser desorption/ionization (SALDI) using the sputtered silver film on glass plate. The SALDI mass spectra were clearly detected as those of the Ag+ adduct forms of [M+Ag]+ without other matrix and cationization agent. The sputtered silver film, which is uniformly deposited on glass, contributes to enhance the ionization and also to supply the cation, Ag+. Therefore, it is expected that SALDI occurs in a good spatial uniformity and the mass image of sample is obtained without the spatial distribution of the material to assist ionization. The silver-coated glass may have a potential to be appropriate in the sample plate for SALDI-IMS. In the presentation, we will discuss on the enhancement of ionization in SALDI using the sputtered silver film, showing the peaks related to the surface plasmon in X-ray photoelectron spectroscopy (XPS).
