Linkage-Specific Sialic Acid Analyses of Plasma N-Glycans in Japanese Semisuper Centenarians

We developed a method for a linkage-specific sialic acid analysis of human plasma N-glycans by a combination of MALDI-TOF MS and sialic acid linkage-specific alkylamidation (SALSA). Using this method, over 100 N-glycans containing multi-branched and highly sialylated were observed in human plasma, and the ratios of α2,3-linked to α2,6-linked sialic acid in two-branched glycans containing fucose were increased in semisuper centenarians compared with those in aged controls.
