Rapid quantitative analysis of enantiomers by supercritical fluid chromatography coupled with triple quadrupole mass spectrometer (SFC-MS/MS) for bioanalysis

Rapid quantitative analysis including chiral separation has been a big challenge for pharmacokinetic studies, where Reversed Phase Liquid Chromatography (RPLC) is commonly used. In this study, we developed and evaluated the high throughput quantitative method for chiral compounds in biological samples using ultra performance Supercritical Fluid Chromatography (SFC) coupled with triple quadrupole mass spectrometry (SFC-MS/MS). As a result, the run time was shortened to 2.5 min whilst maintaining the separation of R- and S-Zopiclone and the data met the criteria of the method validation guideline for Bioanalysis. The ultra performance SFC-MS/MS system offers rapid quantitative analysis of enantiomers in biological samples and it is expected to be a new option for Bioanalysis.
