Optimization of 13C tracer substrates for 13C-based metabolic flux analysis

13C-based metabolic flux analysis (13C-based MFA) is effective approach to quantify metabolic reaction rate in microorganism by using mass spectra data of metabolites. In this study, an application software, Pyflux, which can determine confidence intervals of each metabolic flux faster, was developed. In addition, optimal 13C-substrate for 13C-based MFA of Escherichia coli was searched. In the result, 95% confidence intervals of fluxes through central metabolism were almost narrow when a mixture ratio of [12C] glucose, [1-13C] glucose, and [U-13C] glucose was a ratio of 0: 8: 2. The result also showed that at a ratio of 4: 1: 5, estimation accuracy of anaplerotic pathway was high, although whole accuracy of metabolic fluxes were low. Moreover, optimal labelling patterns of glucose such as [1, 5-13C] glucose, and [1, 2-13C] glucose were indicated.
