Analysis of O-linked Glycan using ASDF-incorporated Curved Field Reflectron: Toward characterization of Biopharmaceuticals

N-/O-linked glycans on glycoprotein play vital roles in biological processes such as stability, protein conformation, intra- and inter- cell signaling, and binding specificity for other biomolecules. In the development of biopharmaceuticals, analysis of these N-/O-linked glycan is very important issue. However, analytical method of O-linked glycan is not standardized yet, whereas the one of N-linked glycan has been improved greatly for the past several years and standardized. The reason of the status is that the optimal enzyme as well as PNGase for N-linked glycans is not existed for O-glycan analysis, and a chemical reaction, which used in general for release of O-glycan, leads to the occasional generation of byproducts. To date, a standardized analytical method of the O-linked glycan has been anticipated. To facilitate the standardization, we applied the new MALDI-tandem TOF to the analysis of the O-glycan and its conjugate with a chemical reaction.
