Facile derivatization for sialyl linkage analysis using solid-phase SALSA
We have developed Sialic Acid Linkage-Specific Alkylamidation (SALSA) for linkage isomer discrimination. A drawback of SALSA is somewhat laborious purification steps to remove excess reagents for the reaction. In this study, SALSA was optimized for solid-phase reaction to facilitate purification steps. This solid-phase SALSA greatly minimized labor and time for derivatizing sialic acids in a linkage-specific manner. We believe that the solid-phase SALSA will be a good choice as a pretreatment of sialyl linkage analysis.
