Evaluation of preparation of recombinant human insulin from Escherichia coli inclusion bodies using TAPS-sulfonate, by nanoLC/ESI electric field FT mass spectrometry
TAPS-sulphonate derivatization enabled recombinant proteins to be prepared efficiently from E. coli inclusion bodies. We evaluated preparation processes of recombinant insulin from TAPS-proinsulin by nanoLC/ESI electric field FT mass spectrometry with use of an LTQ Orbitrap Discovery mass spectrometer. The converted mass value indicated in deconvoluted mass spectrum of TAPS-proinsulin was 5 daltons less than the calculated (monoisotopic) mass value. This error was because of the presence of quaternary ammonium cation in TAPS and of the algorism of the software for deconvolution.
