It is important to understand how different molecular compositions of phosphatidylcholines (PCs) are established in the tissues, cells and organelles, and how the changes in the fatty acid composition of PCs are induced. The three groups of enzymes, phospholipase A2, acyl-CoA synthetase and lysophospholipid acyltransferase are involved in the production of different varieties of phospholipids. Then, free fatty acids (FFAs), the acyl-CoAs and PCs level in the olfactory bulb, cerebral cortex, hippocampus, cerebellum, hypothalamus, midbrain and medulla oblongata were determined by LC-ESI-MS/MS. Each FFAs, acyl-CoAs and PCs of C16:0, C18:0, C18:1, C20:4 and C22:6 were primarily contained in 7 brain areas. On the other hand, we also raise problems of the FFAs, acyl-CoAs and PCs analysis using LC-ESI-MS/MS.
