New On-line Rapid Solid Phase Extraction System for LC/HRMS Structural Analysis using Nonvolatile Mobile Phases

High performance liquid chromatography-mass spectrometry (HPLC-MS) is an effective tool used commonly for structural analysis of impurities in quality control for pharmaceuticals and agrichemicals. However nonvolatile mobile phases which are not appropriate for MS are often used in analytical procedure for these compounds. We have developed an on-line rapid solid phase extraction (SPE) system using two-dimensional liquid chromatography (2D LC) technology, and achieved on-line rapid (within 0.5 minutes) desalting and continuous detection of the elution by mass spectrometry. A total ion current chromatogram (TICC) consistent with the UV chromatogram results in easier identification of target peaks. This new system contributes greatly to the direct structural identification of trace impurities by LC-MS using nonvolatile mobile phases.
