Localization analysis of hormone for rice plant by Nano-particle assisted laser desorption/ionization imaging MS

Plant hormone, acts as a regulator of plant development and adaptation, is an essential transmitter in planta. Mass spectroscopy (MS) technology (e.g. GC-MS and LC-MS) has been well developed to measure plant hormone contents in plant tissue. However, these localizations are unable to determine in the tissue homogenate by the conventional methods. A new quantitative analysis is required to detect localization of plant hormone. Abscisic acid (ABA: Mw264) is involved in plant adaptation under drought, salinity and waterlogging conditions. Reduced soil water and increased salt concentrations stimulate ABA biosynthesis in roots, however waterlogging conditions reduce it1. Interestingly, sometimes ABA signaling is up-regulated under waterlogged conditions although its ABA concentration is reduced in root. We hypothesized that ABA localization in roots is altered under waterlogged conditions and the shift of specific ABA localization initiates ABA signaling. To test this possibility, we used an imaging MS technique2 to determine ABA localization in roots grown in aerated or deoxygenated stagnant agar nutrient solutions (mimicked waterlogged soil conditions). In this research, we revealed the localization of ABA of roots under diverse conditions, visually. ABA localized at root tip under stagnant conditions. On the other hand, ABA uniformly existed in roots under aerated conditions.
